I was prescribed to use upper night guard. After wearing the night guard for 2 nights, I feel that my upper teeth no. 24 is not properly aligning with the lower teeth. Is it possible that the teeth shifts only after 2 nights usage of night guard in case the fitment is not proper

Night guard is prescribed by dentist to prevent the tooth attrition caused by bruxism(tooth grinding).
It will not move the tooth,it will protect the tooth by abnormal habits.
You should visit dentist ask about this and repeat the clinical examination wearing night guard.
In case of occlusal discrepancy,dentist will correct it.
