How many times one should floss his teeth in a day. If it is one time only then when to do it preferably.

Atleast once daily. Every night after dinner. You can use it after every meal if you feel there food wedged between your teeth instead of a toothpick. Be careful not to injure the gums too much

Flossing should be preferably done after each session of toothbrushing. So twice daily schedule will be considered better. Regards.
