The highest concentration of Beta-hcg is in the  first urine of the day (early morning) atleast in the initial weeks. Hence it's essential to do it in the morning. Moreover things just don't confirm after one test,I'd suggest you repeat it for 3 consecutive days. The sensitivity of home pregnancy tests is far less than the Serum Beta-hcg, so it's a better option.

The next step to confirm pregnancy is Serum HCG levels or SOnography. Will also give a good idea about any abnormalities like PCOD. Rest your doctor can advise them well.

Please take beta HCG test to confirm pregnancy.. If negative then other hormonal tests like prolactin, FSHLH, TFT along with AMH and USG pelvis should be done to know the cause of delayed menses.

Blood Hcg will let you know about your pregnancy.
Health Tips
But do not try to go and get the test done on your own. Please get a consult from a physician and let them prescribe this test for you. Doctor is not responsible if the patient is going against medical advise and self medicating.
