Hi,  This is problem with your knee cap, I think according to me,  u are developing retro patellar syndrome, your complaints shows your cartilage at back of patella is at compromised position.  So during knee flex positions it get hurts and as soon you stand up,  it causes fibrillations which causes pain.
Next Steps
if possible a clinical check to confirm diagnosis by expert ortho, till the time take anti inflammatory,  and ice your knee twice a day,  while doin gym put elastic crepe bandage,  physiotherapy for knee muscles
Health Tips
avoid knee flex for more durations,  try to modify your gym exercises where you do standing stuff more.
