Dynamics between 3 of you is needed to be assessed thoroughly to find the exact cause of the issue. As your child is reacting normally to you and not your wife, at child's end, no speech or language disorder seems to be present for now crosssectionaly. It looks like issues in bonding of a child with mother, which usually never occur specialy in Indian setup. Mother is for sure having some signs and symptoms of depression however it's extent can be determined with further detailed assessment for treatment purpose.
Next Steps
Child needs to be assessed for any speech or language disorder though it seems unlikely that he may have it. Dynamics between child and mother and whole family needs to be assessed. 1st consult a child psychiatrist for your child. He might assess any depressive signs in your wife too and would refer her to adult psychiatrist if required. You mentioned your age 43 and child's age around 5 years, so is he the youngest one among your children? Information about other family members and siblings is also must.
Health Tips
In child psychiatry, it takes time for detailed assessment. Sometimes a child psychiatrist can take 4 to 5 sessions only for assessment. So be patient, the solution is definitely going to be there!
