My wife menstrual cycle date was 28th Dec, still she has not got it. She tested with urine test kit and 2 lines were shown. Dark at C level and little darkish line on T level.
Today we went for Beta hcg test and report says 1228 mul/ml.
Does it mean positive pregnancy?

B hcg report says 3 weeks pregnant... it matches

Yes. It means a positive pregnancy but till the bhcg doesn't reach a value of about 1500-2000 it won't show on the ultrasound.
Its also important not to miss an ectopic pregnancy (pregnancy in the fallopian tube) so it's essential you visit a gynecologist.
