Best thing is to ask a home service and let them collect samples . If you want to do it yourself make sure to wear a mask and gloves and then take sample . Yes , can be taken through ear buds

Sampling is done in sterile swabs. And then again placed in media or sterile environment so that any organisms or infections in the environment do not alter the results. Get one of those sample collection vials or visit a professional. And even if ear buds is the only feasible option for you. Then make sure the sample reached the testing institution within 20 minutes

Samples need to be collected sterile and fresh. If you take him to the vet, he will collect it as per procedure. In case your vet asked you to collect, then you need to use sterile swab and collect the sample and reach your vet with in 20.mins.
