When performed properly, this method allows a quantitation of the proportion of mutants resistant to a drug and can detect the 1% proportion of drug- resistant mutants above which therapeutic failure is likely. The present study reports the prevalence of Ofloxacin (OFX) and Kanamycin (KM) resistance in Mycobacterium tuberculosis isolates from cases of pulmonary tuberculosis, who received anti tubercular treatment for >4 weeks in past. As an initial step in evaluating kanamycin DST by MGIT, this study used the MGIT system to determine the MICs for eight kanamycin-susceptible strains (five clinical isolates and three ATCC strains) and four kanamycin-resistant strains. A critical concentration of 5 _g/ml was therefore chosen as the breakpoint for evaluating kanamycin DST by MGIT. This breakpoint was also considered justifiable because 5 _g/ml is the recommended critical concentration for kanamycin susceptibility testing in several media (e.g., BACTEC 7H12 broth).
