I have to take a test on second day of period. So my current cycle is  my period starts in middle of the day (suppose at 6pm) then 6pm of the next day (24hours) will be day one of my cycle. Is it right way to count.

Yes. A cycle is counted from the first day of 1 period to the first day of the next period. Your period started at 6 PM then 6 PM of next day will complete you 1 period day and start the next period day.
